Circular RNA circMYL1 Inhibit Proliferation and Promote Differentiation of Myoblasts by Sponging miR-2400

چکیده

Circular RNAs (circRNAs) are a class of endogenous non-coding (ncRNAs) involved in regulating skeletal muscle development by sponging miRNAs. In this study, we found that the circMYL1 expression was down-regulated during myoblast proliferation, while gradually up-regulated differentiation. The potential role identified proliferation bovine through mRNA and protein marker genes (PCNA, CyclinD1, CDK2), cell counting kit-8 assay, flow cytometry analysis, 5-ethynyl 2′-deoxyuridine (EdU) assay. Analysis differentiation (MyoD, MyoG, MYH2) immunofluorescence Myosin heavy chain (MyHC) used to assess analysis revealed inhibited primary myoblast. Furthermore, demonstrated promoted luciferase screening RNA immunoprecipitation (RIP) assays could have interaction with miR-2400. Additionally, miR-2400 myoblast, may eliminate effects miR-2400, as showed rescue experiments. Together, our results novel circular inhibit promote

Circular RNA hsa_circ_000984 promotes colon cancer growth and metastasis by sponging miR-106b

Circular RNAs (circRNAs) as a novel type of noncoding RNAs (ncRNAs) are widely studied in the development of human various diseases, including cancer. Here, we found circular RNA hsa_circ_000984 encoded by the CDK6 gene was remarkably upregulated in the tissues of colorectal cancer (CRC) patients and in the CRC cell lines. Moreover, high expression level of hsa_circ_000984 was significantly ass...

miR-34s inhibit osteoblast proliferation and differentiation in the mouse by targeting SATB2

A screen of microRNAs preferentially expressed in osteoblasts identified members of the miR-34 family as regulators of osteoblast proliferation and/or differentiation. Osteoblast-specific gain- and loss-of-function experiments performed in vivo revealed that miR-34b and -c affected skeletogenesis during embryonic development, as well as bone mass accrual after birth, through two complementary c...

Circular RNA and miR-7 in cancer.

MicroRNAs (miRNA) play important roles in fine-tuning gene expression and are often deregulated in cancer. The identification of competing endogenous RNA and circular RNA (circRNA) as important regulators of miRNA activity underscores the increasing complexity of ncRNA-mediated regulatory networks. Particularly, the recently identified circular RNA, ciRS-7, which acts as a designated miR-7 inhi...

miR-448 downregulates MPPED2 to promote cancer proliferation and inhibit apoptosis in oral squamous cell carcinoma.

The incidence of oral squamous cell carcinoma (OSCC) is continuously increasing while its survival rate has not notably improved. There is a pressing need for improved understanding of the genetic regulation of OSCC tumorigenesis and progression. In this study, the function of miR-448 in the regulation of OSCC growth and its putative target were thoroughly analyzed in vitro. The expression of m...

Long non-coding RNA MALAT1 acts as a competing endogenous RNA to promote malignant melanoma growth and metastasis by sponging miR-22

Long non-coding RNAs (lncRNAs) are involved in tumorigenesis. Metastasis-associated lung adenocarcinoma transcript 1 (MALAT1), an lncRNAs, is associated with the growth and metastasis of many human tumors, but its biological roles in malignant melanoma remain unclear. In this study, the aberrant up-regulation of MALAT1 was detected in melanoma. We determined that MALAT1 promotes melanoma cells ...
